Acetoanaerobium pronyense sp nov., an anaerobic alkaliphilic bacterium isolated from a carbonate chimney of the Prony Hydrothermal Field (New Caledonia) ArchiMer

A novel anaerobic bacterial strain, ST07-YET, was isolated from a carbonate chimney of the Prony Hydrothermal Field (PHF) in New Caledonia. Cells were Gram-stain-positive, straight rods (0.7-0.8 x 3.0-5.0 mu m) and motile by means of lateral flagella. Strain ST07-YET was mesophilic (optimum 35 degrees C), moderately alkaliphilic and halotolerant (optimum pH 8.7 and 5 gr l(-1) NaCl). Elemental sulfur, sulfate, thiosulfate, sulfite, nitrate and nitrite were not used as terminal electron acceptors. Yeast extract, peptone, tryptone, Casamino acids, crotonate, pyruvate, galactose, maltose, sucrose, ribose, trehalose and glucose were used as carbon sources. Clostridium caminithermale sp nov., a slightly halophilic and moderately thermophilic bacterium isolated from an Atlantic deep-sea hydrothermal chimney ArchiMer

A strictly anaerobic, slightly halophilic and moderately thermophilic, sporulating rod designated strain DVird3(T) was isolated from deep-sea hydrothermal vent samples collected at a depth of approximately 800 m on the Atlantic Ocean Ridge. Strain DVird3(T) possessed a few laterally inserted flagella, had a DNA G+C content of 33.1 mol% and grew optimally at pH 6.6 and at 45 degreesC. Growth was observed at temperatures between 20 and 58 degreesC and at pH values between 5.8 and 8.2. The optimum NaCl concentration for growth was 3% sea salt (30 g l(-1)); no growth was observed in the presence of 15 or 60 g sea salt l(-1). Continuous enrichment culture and molecular monitoring to investigate the microbial diversity of thermophiles inhabiting deep-sea hydrothermal ecosystems ArchiMer

The microflora developing during a continuous enrichment culture from a hydrothermal chimney sample was investigated by molecular methods. The culture was performed in a gas-lift bioreactor under anaerobic conditions, at 90 degrees C and pH 6.5, on a complex medium containing sulfur as the terminal electron acceptor. Archaeal and bacterial diversity was studied. Microorganisms affiliated with the genera Pyrococcus, Marinitoga, and Bacillus were detected through DGGE analysis of 16S rDNA. Additional sequences phylogenetically related to Thermococus and epsilon-Proteobacteria were detected by cloning and sequencing of 16S rDNA from two samples of the enrichment culture. Desulfobotulus mexicanus sp. nov., a novel sulfate-reducing bacterium isolated from the sediment of an alkaline crater lake in Mexico ArchiMer

A novel Gram-negative, non-spore-forming, vibrio-shaped, anaerobic, alkaliphilic, sulfate-reducing bacterium, designated strain PAR22NT, was isolated from sediment samples collected at an alkaline crater lake in Guanajuato (Mexico). Strain PAR22NT grew at temperatures between 15 and 37 °C (optimum, 32 °C), at pH between pH 8.3 and 10.1 (optimum, pH 9.0–9.6), and in the presence of NaCl up to 10 %. Marinitoga hydrogenitolerans sp nov., a novel member of the order Thermotogales isolated from a black smoker chimney on the Mid-Atlantic Ridge ArchiMer

A novel, thermophilic, anaerobic bacterium that is able to tolerate hydrogen was isolated from a deep-sea hydrothermal chimney collected at the Rainbow field on the Mid-Atlantic Ridge. Cells were rod-shaped and surrounded by a sheath-like outer structure (toga); they were weakly motile by means of a polar flagellum. They appeared singly, in pairs or in short chains. They grew at 35-65 degrees C (optimum 60 degrees C), pH 4.5-8.5 (optimum pH 6.0) and 10-65 g sea salts l(-1) (optimum 30-40 g l(-1)). High amounts of hydrogen are emitted in the serpentinite-hosted hydrothermal field of the Prony Bay (PHF, New Caledonia), where high-pH (similar to 11), low-temperature (< 40 degrees C), and low-salinity fluids are discharged in both intertidal and shallow submarine environments. Active carbonate chimneys from the shallow marine serpentinizing Prony Hydrothermal Field were sampled 3 times over a 6 years period at site ST09. Archaeal and bacterial communities composition was investigated using PCR-based methods (clone libraries, Denaturating Gel Gradient Electrophoresis, quantitative PCR) targeting 16S rRNA genes, methyl coenzyme M reductase A and dissimilatory sulfite reductase subunit B genes. Serpentinicella alkaliphila gen. nov., sp nov., a novel alkaliphilic anaerobic bacterium isolated from the serpentinite-hosted Prony hydrothermal field, New Caledonia ArchiMer

A novel anaerobic, alkaliphilic, Gram-stain-positive, spore-forming bacterium was isolated from a carbonaceous hydrothermal chimney in Prony Bay, New Caledonia. This bacterium, designated strain 3b(T), grew at temperatures from 30 to 43 degrees C (optimum 37 degrees C) and at pH between 7.8 and 10.1 (optimum 9.5). Added NaCl was not required for growth (optimum 0-0.2 %, w/v), but was tolerated at up to 4 %. Yeast extract was required for growth. Thermosipho atlanticus sp nov., a novel member of the Thermotogales isolated from a Mid-Atlantic Ridge hydrothermal vent ArchiMer

A novel anaerobic, thermophilic and heterotrophic bacterium, designated strain DV114O(T), was isolated from a deep-sea hydrothermal vent sample from the Mid-Atlantic Ridge. The cells were non-motile straight rods, 1-8 mum long and 0-4 mum wide, surrounded by an outer sheath-like structure (toga). They grew at 45-80 degreesC (optimum 65 degreesC), pH 5-0-9-0 (optimum pH 6-0) and at sea salt concentrations of 20-60 gl(-1) (optimum 30 gl(-1)). Vallitalea guaymasensis gen. nov., sp nov., isolated from marine sediment ArchiMer

A novel obligately anaerobic, non-spore-forming, rod-shaped mesophilic, halophilic, Gram-stain-negative bacterium, was isolated from sediments of Guaymas Basin. The strain, designated Ra1766G1(T), grew at 20-40 degrees C (optimum, 30-35 degrees C) and at pH 6.0-8.0 (optimum, pH 6.5-7.5). It required 0.5-7.5 % NaCl (optimum, 2-3 %) for growth.
